Tina Fey interpreta Liz Lemon, a produtora executiva e escritora do programa de variedades The Girlie Show (Programa das Meninas), um programa fictício, com sketches tal como Saturday Night Live, mas dirigido principalmente ao público feminino. O novo executivo do canal Jack Donaghy, interpretado por Alec Baldwin, recém-contratado, recorre às suas tácticas de marketing para aumentar as audiências do programa, e decide contratar Tracy Jordan (Tracy Morgan), uma estrela de cinema (com uma carreira baseada em filmes protagonizados por Martin Lawrence, Eddie Murphy e Damon Wayans e cuja personalidade não difere muito da do próprio Tracy Morgan) para fazer parte do elenco. Jane Krakowski interpreta Jenna Maroney, a estrela do The Girlie Show, que passou a se chamar TGS with Tracy Jordan, para melhor destacar a participação de Jordan.
30 Rock é um sucesso de crítica, recebendo vários prêmios (incluindo três prêmios Emmys consecutivos de melhor seriado de comédia). Em 2009, foi recordista de indicações (22 no total) ao Emmy.
